Your question is not clear.

If you're asking which program in a transaction is using a file, use CEDF to trace the CICS calls until the file is accessed, then the current program name is part of the display.

If you're asking generically how to tell which programs use which files in a CICS region, the only answer I am aware of is manual code inspection. The file name can be a variable, so only looking at the code will let you know for sure which programs in a region use a given file.
